ニキビ菌に関する報告書

ただいま、タイにおきまして臨床実験中です。

Method 1

1. prepare BHA agar in DI water and sterilize with autoclave for 20 min.
2. add glucose in BHA to have final concentration 1%
3. pour agar base in each plate to be 15 mL, leave until it get harden
4. spread sample (spray 0.5 or 1.0 mL) per plate
5. prepare P.acnes at MCF = 0.5 (number colony = 10⁸ CFU/mL)
6. prepare P.acnes at varying dilution 10^-1, 10^-2 , 10^-3 , 10^-4 , 10^-5 , 10^-6
7. mix each dilution of P.acnes 100 uL of 10^-5 , 10^-6 with 4.5 mL BHA broth
8. pour broth on the plate and leave until it become solid
9. incubate at anaerobic condition at 37 °C in anaerobic jar for 3 days

Result

* = no P.acnes growth
Positive control = clinda M ^®

Note

• pray (for both 0.5 and 1 mL) can reduce the growth of P.acnes at all dilutions although the inhibition ability is lower than positive control .
• Higher amount of spray (1 mL) can inhibit the growth of microbe better than lower volume.

Method 2

1. prepare BHA agar in DI water and sterilize with autoclace for 20 min.
2. add glucose in BHA to have final concentration 1%
3. pour agar base in each plate to be 15 mL, leave until it get harden
4. prepare BHA seed agar 4 mL and sample (spray 1.0 mL) per plate
5. pour BHA seed agar on top of base agar, leave until harden
6. prepare P.acnes at MCF = 0.5 (number colony = 10⁸ CFU/mL)
7. prepare P.acnes at varying dilution 10^-1, 10^-2 , 10^-3 , 10^-4 , 10^-5 , 10^-6
8. spread dilution of P.acnes 100 uL of 10^-4 , 10^-5 , 10^-6 dilution
9. incubate at anaerobic condition at 37 °C in anaerobic jar for 3 days

Result

* = no P.acnes growth
Positive control = clinda M ^®

Note

• Spary can reduce the growth of P.acnes as compared to negative control without any treatment. However the inhibition ability towards microbe is still lower than positive control (clindamycin)
